Ziel der vorliegenden Studie war es, ein thermoreversibles, am Auge in situ bildendes Hydrogel auf Chloramphenicol-Basis zu entwickeln und zu bewerten, um die Verweildauer auf der Hornhautoberfläche zu verlängern und die Bioverfügbarkeit am Auge zu verbessern. Die Formulierung wurde unter Verwendung von Poloxamer 407 als thermosensitivem Gelierpolymer sowie von HPMC oder Carbopol 940 als Viskositätsverstärker hergestellt. Die hergestellten Formulierungen wurden hinsichtlich Aussehen, pH-Wert, rheologischem Verhalten, Gelierungstemperatur, Wirkstoffgehalt, In-vitro-Wirkstofffreisetzung, antimikrobieller Aktivität und Stabilität charakterisiert. Unter den entwickelten Formulierungen wies die Charge F4, die 12 % Poloxamer 407 und 0,5 % HPMC enthielt, optimale physikalisch-chemische Eigenschaften, ein geeignetes Gelierverhalten, eine verzögerte Wirkstofffreisetzung sowie eine wirksame antimikrobielle Aktivität auf. Diese Ergebnisse deuten darauf hin, dass das entwickelte In-situ-Hydrogel ein vielversprechendes ophthalmologisches Arzneimittelabgabesystem für eine verlängerte therapeutische Wirksamkeit von Chloramphenicol darstellt.
