La présente étude avait pour objectif de mettre au point et d’évaluer un hydrogel oculaire in situ thermoréversible à base de chloramphénicol afin d’augmenter la durée de séjour précornéenne et d’améliorer la biodisponibilité oculaire. La formulation a été préparée à partir de Poloxamer 407 comme polymère gélifiant thermosensible, avec de l’HPMC ou du Carbopol 940 comme agents augmentant la viscosité. Les formulations préparées ont été caractérisées en fonction de leur aspect, de leur pH, de leur comportement rhéologique, de leur température de gélification, de leur teneur en principe actif, de la libération in vitro du principe actif, de leur activité antimicrobienne et de leur stabilité. Parmi les formulations développées, le lot F4, contenant 12 % de Poloxamer 407 et 0,5 % d’HPMC, a présenté des propriétés physico-chimiques optimales, un comportement de gélification approprié, une libération prolongée du principe actif et une activité antimicrobienne efficace. Ces résultats indiquent que l’hydrogel in situ mis au point constitue un système prometteur d’administration ophtalmique permettant une efficacité thérapeutique prolongée du chloramphénicol.
